Isolation of urine subcellular particles

S-TECH is used to isolate subcellular particles using our track-etched polycarbonate membranes. The track-etched pores are highly uniform, and the membranes are low-binding because there is no traditional fibre membrane matrix to entrap particles. They are ideal for isolating subcellular particles such as exosomes and microvesicles. They also concentrate; they enable the removal of excess fluid and the recovery of particles for the next stage of their analysis. Buffer exchange is also easy using the S-TECH system, enabling the transfer of the recovered particles into the most appropriate reagent mix for secondary analysis.

When using S-TECH polycarbonate track etch membranes, it is important to prevent the sample from drying on the membrane.

Isolation of serum subcellular particles

Attached is a video demonstrating S-TECH's versatility in isolating subcellular particles from human serum. Our EV isolation membranes use geometrically precise track-etched pores in thin-film polycarbonate, a configuration that eliminates the entrapment of subcellular particles that occurs with fibre- or mesh-type filtration systems. In the video, we demonstrate how easy S-TECH is to use as a single device for the isolation, purification, concentration and buffer exchange of subcellular particles from 125 µL of serum (250ul total volume diluted in PBS 1:1). We also demonstrate the particle size distribution by tunable resistive pulse sensing (TRPS) and the efficient protein purification that occurs during the S-TECH processing of the serum sample.
